In 2008, roughly 14.3 million Americans were taking antipsychotics?typically prescribed for bipolar disorder, schizophrenia, or a number of other behavioral disorders?making them among the most prescribed drugs in the U.S. Almost all of these medications are known to cause the metabolic side effects of obesity and diabetes, leaving patients with a difficult choice between improving their mental health and damaging their physical health. In a paper published January 31 in the journal Molecular Psychiatry, researchers at Sanford-Burnham Medical Research Institute (Sanford-Burnham) reveal how antipsychotic drugs interfere with normal metabolism by activating a protein called SMAD3, an important part of the transforming growth factor beta (TGFbeta) pathway.

The TGFbeta pathway is a cellular mechanism that regulates many biological processes, including cell growth, inflammation, and insulin signaling. In this study, all antipsychotics that cause metabolic side effects activated SMAD3, while antipsychotics free from these side effects did not. What's more, SMAD3 activation by antipsychotics was completely independent from their neurological effects, raising the possibility that antipsychotics could be designed that retain beneficial therapeutic effects in the brain, but lack the negative metabolic side effects.

"We now believe that many antipsychotics cause obesity and diabetes because they trigger the TGFbeta pathway. Of all the drugs we tested, the only two that didn't activate the pathway were the ones that are known not to cause metabolic side effects," said Fred Levine, M.D., Ph.D., director of the Sanford Children's Health Research Center at Sanford-Burnham and senior author of the study.

In a previous study aimed at developing new insights into diabetes, Dr. Levine and his team used Sanford-Burnham's high-throughput screening capabilities to search a collection of known drugs for those that alter the body's ability to generate insulin, the pancreatic hormone that helps regulate glucose. That's when they first noticed that many antipsychotics alter the activity of the insulin gene. In this current study, the researchers set out to connect the dots between antipsychotics and insulin. In doing so, experiments in laboratory cell-lines showed that antipsychotics known to cause metabolic side effects also activated the TGFbeta pathway?a mechanism that controls many cellular functions, including the production of insulin?while the drugs without these side effects did not.

Wondering whether their initial laboratory observations were relevant to the human experience, the researchers reanalyzed previously published gene expression patterns in brain tissue from schizophrenic patients treated with antipsychotics. What they found supported their earlier findings?TGFbeta signaling was activated only in those patients receiving antipsychotic treatment. Looking further, they found that the extent to which each antipsychotic drug activated the TGFbeta pathway in human brains correlated very closely with the extent to which those same drugs activated SMAD3 and affected the insulin promoter in their cell culture experiments.

The TGFbeta pathway also plays an important role in metabolic disease in people who don't take antipsychotic medications. "It's known that people who have elevated TGFbeta levels are more prone to diabetes. So having a dysregulated TGFbeta pathway?whether caused by antipsychotics or through some other mechanism?is clearly a very bad thing," said Dr. Levine. "The fact that antipsychotics activate this pathway should be a big concern to pharmaceutical companies. We hope this new information will lead to the development of improved drugs."

The Dow Jones industrial average rose 3.4 percent in January and the Standard & Poor 500 gained 4.4 percent, the best performances for both indexes to open a year since 1997.

In what was mostly a slow and steady climb, the Dow Jones industrial average rose 3.4 percent in January and the Standard & Poor's 500 gained 4.4 percent, the best performances for both indexes to open a year since 1997.

Investors were encouraged by modest but welcome improvement in the U.S. economy, including an 8.5 percent unemployment rate, the lowest in almost three years. Corporate profits didn't wow anyone ? except Apple's ? but they were good enough.

"I don't see anything really glamorous or tremendous about the economy or earnings," said Jerry Harris, chief investment strategist at the brokerage Sterne Agee. "But I think they're very acceptable, and things are grinding along."

An unexpected drop in consumer confidence dragged stocks down on the final day of the month. The Dow Jones industrial average finished down 20.81 points, or 0.2 percent, at 12,632.91.

The broader market fared better. The S&P barely finished in the red, declining 0.60 point to 1,312.41. The Nasdaq composite index rose 1.90 points to close at 2,813.84. The Nasdaq gained 8 percent for the month, its best January since 2001.

In January 1997, the last time stocks had such a fast start, the S&P gained 6.1 percent. Bill Clinton was inaugurated for his second term. An Asian financial crisis and "Titanic" lay ahead. Later that year, the Dow crossed 7,000 and 8,000 for the first time.

This January, analysts said, investors had such low expectations for the economy that it was easy for things to turn out better than expected.

"There are no big surprises," said Kim Caughey Forrest, a senior equity analyst at money manager Fort Capital Group. "That's the kind of ho-hum economy that we are in right now."

The Dow closed at 12,217.56 at the end of last year, then started this year with a pop ? a gain of 179.82 points on opening day. It was the kind of big swing investors became accustomed to in 2011.

Since then, it's been a quiet ascent: 19 days in a row of moves of less than 100 points. The last time the Dow had such a placid stretch was a 34-day run that started Dec. 3, 2010.

Scottrade, the online brokerage, said stock buyers outpaced sellers among its clients for the first 14 trading days of the year, Jan. 3 to Jan. 23. It also said volume was 16 percent higher than December's average.

On Tuesday, the Dow started up 66 points after encouraging signs from Europe that Greece might finally complete a deal to cut its crushing debt, a step toward securing a critical ?130 billion bailout payment.

Greece is negotiating with investors who bought its government bonds. They are expected to swap their bonds for new ones with half the face value, plus a lower interest rate and longer term of maturity.

Investors are increasingly worried that Portugal may need a similar deal with its private creditors. European leaders insist the Greek reduction is a one-time event. Portugal's borrowing costs have risen to record highs.

The Dow lost its gains after consumer confidence fell to 61.1 in January, down from 64.8 in December. Economists had expected 68. The Conference Board said Americans are more worried about their incomes, gas prices and business conditions.

There were also signs that the housing market continues to struggle. Home prices fell in November for a third straight month in in 19 of the 20 cities tracked by the S&P/Case-Shiller index. The biggest declines were in Atlanta, Chicago and Detroit.

In the commodities market, investors worried that the confidence figure was a sign of weaker demand to come, and they sold industrial metals that have prices closely tied to the economy.

Copper for March delivery dropped 3.65 cents to $3.79 per pound, and March palladium ended down $2.15 at $686.35 per ounce. April platinum fell $28.20 to $1,588.10 an ounce.

The metals ended the day down after wild swings. Traders bid up prices in morning trading, encouraged by news that European officials were making progress to contain the financial crisis there, then sold hard on the confidence number.

"This is a day that every trader takes Tums," said George Gero, vice president at RBC Global Futures.

Precious metal prices ended the day mixed. The price of gold rose, as it often does when it looks like the economy might shrink or the dollar might lose its value. Gold for April delivery gained $6 to finish at $1,740.40 an ounce.

In the bond market, the weak U.S. economic data and uncertainty about Greece lit up demand for safe investments. The benchmark 10-year Treasury yield dipped to 1.795 percent, its lowest close in almost four months.

The yield on the five-year Treasury note hit a record low for the second straight day, falling to 0.70 percent.

Treasury yields have been falling since last week, when the Federal Reserve said it expected to hold interest rates near zero into late 2014, more than a year longer than its last estimate, because the economic recovery will need help.

In corporate news:

? RadioShack Corp. stock plummeted 30 percent after the company said its profit fell sharply ? 11 cents to 13 cents per share for the quarter that ended in December, down from 51 cents a year earlier and less than half what Wall Street was expecting.

? Best Buy Co. Inc., one of RadioShack's competitors, responded by falling 5.6 percent, worst in the S&P. Both companies sell and service cellphones, but demand has softened at their stores.

? Avery Dennison Corp., which makes labels and packaging materials, fell 5.6 percent after it said earnings plunged 81 percent on nearly flat sales. Its 2012 outlook was well below Wall Street expectations.

? Mattel Inc. soared 5 percent because of strong demand for Barbie and Monster High dolls during the holidays. That boosted Mattel's fourth-quarter profit by a better-than-expected 14 percent. The company also raised its dividend.

? U.S. Steel Corp. gained 5 percent after it reported strong demand for pipes from the oil industry from October through December. The company was also optimistic about this quarter.

? Agriculture conglomerate Archer Daniels Midland declined 3.6 percent after it reported an 89 percent drop in quarterly net income. The company said its results were weighed down by weakness in oilseeds, corn processing and agricultural services.

Shortly before midnight, our pilot Don Glaser shakes my sleeping bag. The sky has dumped something halfway between snow and rain, and it is freezing to the wings of the Flagship.

We all head to the harbor, wrestling with a northeast wind as we try to stretch nylon covers over the wings. Slush has also begun to bubble up through a nearby crack in the ice. We fear the Flagship?s skis will freeze into the slush, so we prop them up on 4x4s. Then it?s back to bed.

It snows all day. So we occupy ourselves with chores around the cabin: hauling water and setting up a sauna. We also prepare the Red Dragon ? a 25-pound propane tank, 12-volt battery, spark-ignited torch and blower, and a hose, all strapped to a toboggan. As soon as the wet snow stops falling, we?ll need that.

Tuesday, Jan. 24

By early morning, the snow has stopped. The temperature drops, and the northeast wind comes up again. After a bowl of oatmeal by the light of our headlamps, we slog to the harbor. Removing the wing covers, we can see that we put them on too late. Every flying surface of the Flagship is coated with ice. Planes fly because their wings are shaped like a foil. Ice distorts that shape and compromises lift. All the ice will have to be removed.

Luckily, that?s what the Red Dragon does best. We haul the contraption onto the icy harbor next to the plane. Firing up the Red Dragon, we heat the flying surfaces, then brush the ice off as it melts. Ice removal is a delicate affair. The Flagship?s wings are wood covered with fabric. Brush at the ice too hard, and the fabric will tear. Heat it too much, and it will catch fire. For three hours, we gently melt, rub and flake off ice.

Our work, observing wolves and moose, depends on the weather. Waiting for snow or wind to stop is routine. Fixing cold-stricken equipment is standard. Wishing it had been just a few degrees colder, so that wet snow hadn?t frozen to the wings: we make lots of ?if only? weather wishes. But the weather is part of what makes a place, any place, special. And as the weather changes, the wolves and moose have much more at stake than we do. Fighting the weather is futile. Appreciating it, even when it?s not pleasant, is as good as it gets.

By noon, the plane is ready for flight, and by 3 p.m., the wind has lightened up. Rolf Peterson, a co-researcher, and Don depart to look for wolves. They find Chippewa Harbor Pack near the site where we?d seen them a couple days ago. It is clear what kept them there: the carcass of a moose they?d killed. The few remaining bones suggest they?d killed it about five days ago, perhaps the day we arrived. Rolf and Don also find fresh tracks of a lone wolf near Halloran Lake, leaving and then returning to a cedar swamp filled with tracks of the snowshoe hares that probably became the wolf?s dinner.

Rolf and Don spend most of the flight searching for Romeo, a radio-collared male from Chippewa Harbor Pack. Flying at 4,000 feet over the island they should pick up any signal. Static is all they hear.

Wednesday, Jan. 25

I get up at 4 a.m. to fire up the portable generator we use to heat the plane?s engine. I might as well have stayed in my sleeping bag. By 6:30 a.m., the clouds had lowered until they touched the island. Freezing drizzle was threatening, so we put the wing covers on again. Why does the weather keep frustrating our desire to collect information?

In the five days that we?ve been here, we?ve made just two research flights. Actually, that?s a pretty typical rate of being grounded. And on each flight, we?ve gained only fragments of insight. But multiply each observation by the 30 to 40 flights we make each winter, then multiply that by more than five decades. It adds up to a significant bank of data.

I spend each day searching for something significant. During each flight, we record the locations of wolves or their tracks. I have condensed several decades of wolf travel routes onto a single map. We are working to quantify what we?ve long intuited: that the predation risk for moose is greatest near the island?s shoreline. It is easier for wolves to walk along shorelines, where the snow is windswept, and moose are attracted to shoreline habitats because they tend to find better forage there.

Knowing that predation risk decreases with distance from the shoreline is also just a tiny insight. Add this to several other long-term observations and some sophisticated analysis from Robert Montgomery, a habitat expert from Michigan State University, put it all together, and, well, our insight is not in perfect focus yet, but it?s looking as though younger, healthier moose spend more time in these risky shoreline habitats during severe winters than do the aging moose who would be more vulnerable to predation.

It feels good to pore through old maps, recalling individual wolves, many of which are now dead, their travels and lives, and the adventures we had making those observations.
